Japanese RCT (Mori et al.) showed cognitive improvement in mild cognitive impairment after 16 weeks. Small studies support mood, peripheral neuropathy recovery, and subjective focus / clarity. Animal evidence for NGF is strong; human-CNS evidence is still emerging. Effects: 4–8 weeks subjective, 12–16 weeks for objective cognitive measures.

Lion's mane is an edible mushroom that contains unique compounds (hericenones in the fruiting body, erinacines in the mycelium) that cross the blood-brain barrier and stimulate Nerve Growth Factor synthesis. NGF supports the maintenance, growth, and regeneration of neurons — making lion's mane one of the very few supplements with a plausible mechanism for actual nerve repair. Human trials have shown cognitive improvements in mild cognitive impairment, modest mood benefits, and notable benefit in peripheral neuropathy and post-injury nerve recovery. It's also a culinary mushroom — meaty texture, edible at standard meal portions.

Form
Dual-extract (water + alcohol) from fruiting body — the most evidence-aligned form. Fruiting body extract standardised to beta-glucans (>20%) is the next best. Mycelium-on-grain is cheaper but contains substantial filler grain; choose only if labelled with verified beta-glucan content. Look for third-party tested products.
Dose
500–1,000 mg of fruiting body extract, 1–3x daily (1,000–3,000 mg/day total). Higher end for cognitive / nerve indications.
Timing
AM with food. Splitting doses (AM + lunch) gives more sustained effects than single dose.
Schedule
Daily. Some users cycle (5 days on, 2 off, or 4 weeks on / 1 off) but evidence for needing this is thin.
Side effects
Generally very well-tolerated. Rarely: itchy skin, GI upset, mild contact dermatitis (in mushroom-allergic individuals).
Interactions
Anticoagulants — mild antiplatelet effect at high doses (clinical relevance small). Generally clean.
Contraindications
Mushroom allergy. Bleeding disorders (caution at high doses).
Lab markers
Not standard. Functional cognitive testing (MoCA, computerised batteries) for objective tracking.
Food sources
The mushroom itself, eaten fresh or cooked — meaty white pom-pom-shaped mushroom, increasingly available at farmers' markets and Asian groceries. Excellent in stir-fries and as a meat substitute. Eating it as food provides modest benefit; therapeutic doses require concentrated extract.
